Dipotassium glycyrrhizinate (GK2) is known as an anionic surfactant having anti-allergic and anti-inflammatory activities. However, physicochemical properties of mixed micelle formation of GK2 is not well-understood. In this study, in order to understand the microenvironmental polarity around the enone chromophore of chiral GK2 and its 20 β-carboxyl methyl ester (GMe) in the presence of long-chain alkyltrimethylammonium bromide (Cn-TAB) homologues, circular dichroism (CD) measurements were carried out. The spectro-scopy revealed that the ability of mixed micelle formation of Cn-TABs with GK2 depended upon the alkyl chain length. C12-TAB and C14-TAB, which have almost the same alkyl chain length as the long axis of the hydrophobic part of GK2, formed the most stable micelles with both GK2 and GMe in the molar ratios of 2 : 1 and 1 : 1, respectively. Surface tension measurements and fluorescence analyses using pyrene as a probe also supported the above results stoichiometrically. In addition, molecular dynamic calculations were in good agreement with those results.
