温度勾配を付与した内径9.7mm, 高さ1430mmの精留塔を装着した超臨界CO<SUB>2</SUB>抽出装置を用い, EPA-Etの高純度精製を検討した。本研究では, 精留効果の発現に必要な内部還流に焦点を当て, 精留塔の充填物の効果, 温度勾配による還流比の違いを考察した。そして, 塔内にDixon Packingを充填して, 塔底から塔頂にかけて35℃～80℃の直線上の温度勾配を付与することで, 分離能を改善し, 抽出油中のEPA含量を約90%以上とした。
Ethyl icosapentaenoic acid (EPA-Et) was concentrated from fish oil with Supercritical CO<SUB>2</SUB> using a rectification column in 9.7 mm internal diameter and 1100 mm effective height. To improve supercritical fluid-liquid contact, "Tawashi" rotor was placed in the extraction cell and the column was packed with Dixon Packing. For greater separation capacity, the column temperature gradient from bottom to top was 35 to 80°C. As the result, reflux flow, waterfall-like in appearance, could be seen, and EPA attained a maximum 90%, although C<SUB>20 : 4</SUB> components were abundantly present in the experimental feedstock. It is thus evident from the present study that EPA can be separated from C<SUB>20 : 4</SUB> components by Supercritical CO<SUB>2</SUB> extraction using rectification column optimized with supercritical CO<SUB>2</SUB>-Liquid contact and minimal backmixing.
