Chiral phase HPLC is a simple, sensitive and accurate method for determining the stereoconfigurations of chiral lipid components. Enantiomers and diastereomers of glycerolipids having one or two hydroxy groups in their molecules, such as diacylglycerols (DG), alkylglycerols and phosphatidylglycerols, have been successfully separated after conversion to the 3, 5-dinitrophenylurethane derivatives by HPLC on chiral stationary phases possessing a naphthyl group. Although the chromatographic resolution of enantiomeric triacylglycerols (TG) has not been carried out to date, it is possible to determine the DG moieties of TG by examining the enantiomeric sn-1, 2- and sn-2, 3-DG generated from TG by partial Grignard degradation. Chiral phase HPLC in conjunction with polar capillary GC, reversed-phase HPLC or mass spectrometry accurately indicates enantiomer compositions of natural DG. This method has been used to determine the positional distribution of aryl groups in TG molecules, positional placement of alkyl groups in glycerol ether molecules and stereospecificity of lipid related enzymes such as lipases.
