As part of basic studies on the mechanism of water purification by aquatic microorganisms, biodegradation of fenitrothion (MEP), diazinon, simazine and aniline were examined using Tempaku river surface water located in the east of Nagoya City, furthermore, changes of the microflora and MEP biodegradability of these isolates in culture media of MEP degradation test by the River Die-away (RDA) method were investigated. Biodegradability of river water showed relatively high to MEP and low to diazinon and simazine. Analyses of the microflora in the river water revealed the presence of bactera of the following genera which are believed to be involved in decomposition of environmental pollutants; Acinetobacter sp., Pseudomonas sp., Alcaligenes sp., Flavobacterium sp. and Molaxella-Brucella sp.. Growth of bactera of these genera were confirmed in culture media of MEP degradation test by the RDA method, and many of the peacies tested showed high MEP degradation acivities. Analyses of products obtained in MEP degradation media by the Handai method and a modified Handai method for isolates showed the presence of aminofenitrothion, but it was not detected in the media by the RDA method. MEP was enzymatically degraded with cell-free extracts to produce aminofenitrothion in the presence of both NADH and NADPH under anaerobic condition. It is considered that reduction of the nitro group in the molecule to form amino compound may be one of the main metabolic pathway of MEP for a certain bacterium.
