Morphological analysis of isolated colonic crypts in rats postnatally indicated that Self-reproduction of crypts occured by a fission mechanism and the division began at the crypt base and proceeded upwards until there were two separate crypts. Occasionally, before the separation was complete, a second fission process started on one or both sides of a bifurcating crypt, and a triple-branched or quadruple-branched crypt resulted. Analysis of isolated aberrant crypt foci (ACF) in rats which were treated with 1, 2-dimethylhydrazine (DMH) revealed that the development of ACF consisting of multiple crypts was also due to a fission mechanism. Initially, an indentation appeared at the base of a single crypt of ACF, followed by formation of a bifurcation and crypt division.<BR>As a result of the sequential analysis of ACF, the number of ACF increased to 20 weeks and decreased from 20 weeks to 30 weeks in this study. The pit patterns of ACF were simultaneously observed. Almost all ACF showed large round pits. However, as time passes, irregular forms, for example, small round or gyrus-like pits, were found occasionally. The crypt base was present in the area where the functional stem cells were found. The level of initiation of stem cells in ACF, which did not demonstrate irregular pattern, was considered to be, too poor to ensure persistence so that they are reversible in nature. Only a few ACF crypts demonstrated irregular pattern in the present study, but they could be regarded as more atypical with a great possibility of developing into tumors.
