Abstract

The activities of enzymes involved in taurine biosynthesis in fish were determined. Precursors of taurine (cysteinesulfinate, cysteate and cysteamine) were incubated with liver homogenate in the presence of various cofactors. Levels of hypotaurine and taurine resulting from enzyme reactions were analyzed by means of OPA-prelabeling and reverse-phase HPLC. Among the fish examined, bluegill, Lepomis macrochirus synthesized significant amounts of hypotaurine and taurine from cysteinesulfinate and cysteate, respectively, although the production rate of hypotaurine derived from cysteinesulfinate was substantially higher than that of taurine from cysteate. Considerable hypotaurine formation from cysteinesulfinate was also observed in rainbow trout, Onchohynchus mykiss. In smelt fish, Plecolossus alives and red sea bream, Pagrus major, cysteamine was readily converted into hypotaurine. Furthermore, red sea bream efficiently converted cysteate into taurine; the formation of which was inhibited by addition of cysteinesulfinate in a dose-depen-dent manner. In the livers of carp, Cyprinus carpio, Japanese flounder, Paralichtys olivaceus, and yellowtail, Seriola quinqueradiata, however, no significant enzyme activity was observed. These data suggest interspecific differences in both the pathway and capacity of taurine biosynthesis in fish.
