We examined the expression of inducible nitric oxide synthase (iNOS) in 63 patients with laryngeal squamous cell carcinoma (SCC) who had undergone treatment at our hospital. The iNOS expression was detected by an immunohistologic method using mouse monoclonal antibody against iNOS and the staining intensity was scored. The iNOS expression in cytoplasm of tumor cells was detected in 36 (57.1%) of the 63 laryngeal cancers. The iNOS expression was not correlated with either arising sites, TN clinical classifications, or stages. In early-stage (T1N0, T2N0) diseases treated with radiotherapy alone, patients who experienced local recurrence after complete remission showed significantly higher iNOS scores than patients who did not experience local recurrence (5.0 vs. 2.0, p=0.0053). The disease-free survival rate of patients with iNOS expressions was significantly poorer than that of patients without iNOS expressions. These findings suggest that the iNOS expression may be useful as a factor for predicting local recurrence and prognosis in early-stage laryngeal SCC.
