The objective of this study was to determine effects of non-steroidal aromatase inhibitor (AI, Fadrozole, Ciba-Geigy) on gonadal differentiation in broiler embryos and broiler chickens. At day 5 of incubation fertilized eggs (of hens) were injected with AI. AI-treated eggs were incubated and sex-reversal effects and levels of P 45017α-hydroxylase (P 450C17) and P 450aromatase (P 450arom) mRNA were evaluated by observation of gonadal phenotype and by Northern and slot blot analysis either at day 18 of incubation or at 2 months of age. Individual genetic sex was evaluated by Southern hybridization analysis using a W-specific DNA probe. Saline injection had no sex-reversal effect, whereas AT injection resulted in 100% and 60% sex-reversal of genetic females in the embryos and chickens, respectively. There was no appreciable sex-reversal effect in genetic males. At day 18, in the saline group, P 450C17 mRNA levels in the ovary were higher than in the testis but, testicular mRNA levels and those in the sex-reversed testis in the AI group were lower than the testicular levels of the control group. P 450arom mRNA levels were higher in the ovary than in the testis in the control group but the levels of the testis and sex-reversed testis in the AI group were lower than those in the ovary. Body weight of sex-reversed females was lower than males at 5, 7 and 8 weeks but was not significantly different from that of normal females at 8 weeks. Plasma concentrations of testosterone of sex-reversed females in chickens showed intermediate levels between those of normal females and males, whereas there was no difference in plasma estradiol levels between the sex-reversed females and normal chickens. P 450C17 mRNA levels in the ovary and testis were high in the control group, but the ovarian and testicular levels in the AI group were lower than those levels of respective gonads of the control group. On the other hand, P 450arom levels in the ovary were significantly higher than those in the testis in the control group but the ovarian and testicular levels in the AI group were lower than those levels of respective gonads of the control group. Those of the sex-reversed testis were significantly lower than those of the normal ovary. This study demonstrated that AI-treated females exhibited suppressed mRNA levels of P 450arom in sex-reversed testis without associated improvement in body weight gain in the broiler chickens.
