Several hybridomas producing monoclonal antibodies specific for Streptococcus rattus (S. rattus) were prepared by fusing myeloma cells with the spleen cells immunised with S. rattus strain FA1. One of the hybridomas was injected into BALB/c mice to produce ascitic fluids from which monoclonal antibody 2-27 (MAb 2-27, IgM) was partially purified after precipitation with ammonium sulphate. Reactivity of MAb 2-27 was examined in EIA against various strains of mutans streptococci as well as other streptococcal species. It was shown that MAb 2-27 reacted only with S. rattus strains and not with any other strain of the mutans group nor with other streptococcal strains tested. Galactosamine, glucosamine and N-acetylgalactosamine strongly inhibited the reaction of the monoclonal antibody with whole cells of S. rattus and a polysaccharide cell wall antigen purified from S. rattus strains FA1 and BHT. Immunoelectron microscopy was conducted to demonstrate distribution and localization of the specific antigen on S. rattus cells by use of MAb 2-27 plus gold-conjugated antibody. It was shown that the antigen determinant was irregularly localized on thread-like projections on the surface of the cell wall.
