To enrich our knowledge of the effects of therapeutic radiation on immunity organs located out of the irradiation field for oral cancer patients, we designed a study to examine the effects of local irradiation on the intestinal mucosal immune systems of laboratory animals. Mouse maxillae were exposed to a dose of 10Gy and the ratios of subsets of intestinal intraepithelial lymphocytes (i-IELs), and Peyer's patches lymphocytes and splenocytes were measured using flow cytometry. The following results were obtained: 1) In intestinal intraepithelial, the percentage of T cells, CD4+ SP T cells, CD8+ SP T cells, CD8+ αα T cells, or CD8+ αβ T cells did not show any significant change after irradiation; 2) In Peyer's patch, the percentage of T cells, CD4+ SP T cells and CD8+ SP T cells did not show any significant change after irradiation; 3) In intestinal intraepithelial and Peyer's patch, the percentage of L-selectin- T cells and L-selectin+ T cells did not show any significant change after irradiation; 4) In the spleen, the percentage of L-selectin- T cells showed a significant increase one day after irradiation, and one of the L-selectin+ T cells showed a significant decrease one day after irradiation. The above findings demonstrate that irradiation does not cause any significant changes in the ratios of subsets of i-IELs and Peyer's patch lymphocytes. From these findings we could conclude that irradiation to the head and neck region has little influence on the intestinal mucosal immune system, and it may suggest that a lymphocyte homing phenomenon affects the ratio of the lymphocyte subset in each immunity organ.
