Relationships among orotate phosphoribosyl transferase (OPRT) protein expression, measured using antihuman OPRT rabbit polyclonal antibody ; OPRT enzyme activity, clinicopathological background factors, and single nucleotide polymorphisms (SNPs) in the OPRT coding region were examined. Thirty-five subjects who had undergone surgical resection of colon cancer in our hospital between December 2003 and July 2004 were enrolled in the study. OPRT activity at the tumor sites was significantly higher than in normal regions. Based on immunohistochemical staining, strong expression of OPRT was noted in 24 subjects. The OPRT activity in these subjects was significantly higher compaired with subjects with low OPRT expression. No relationships were found among OPRT protein expression, clinicopathological background factors, and SNPs in the OPRT gene. These results suggest that OPRT enzyme activity can be predicted using OPRT immunohistochemical staining, and that patients with hypersensitivity to 5-fluorouracil (5-FU) can easily be identified using this method.
