Abstract

キノコに含まれるACE阻害物質について,野生,栽培種,市販品の食用キノコ計23種を調査した.熱水抽出物のACE阻害活性を調べた結果,シメジモドキ,スミゾメシメジ,ホウキタケ,コウタケなどのキノコに強い阻害活性が認められた.各キノコの抽出固形物のIC<SUB>50</SUB>は,ヌメリスギタケ(野生種,栽培品),コウタケ,ホウキタケ,スミゾメシメジの順で強いことが示された.透析膜による分画では,キノコの種類によりACE阻害を示す物質に特徴があることが示された.<BR>シメジモドキの70%エタノール抽出画分を種々のクロマトグラフィーによる分画を行った結果,ACE阻害物質が単離され,ニコチアナミンであると同定された.シメジモドキに含まれるニコチアナミン量は13.2mg/dry 100gであった.<BR>また,他の23種のキノコについて,ニコチアナミンの有無を調査した結果,ホウキタケに28.7mg/dry 100g含まれていた.イッポンシメジ科ではシメジモドキ以外には含有が認められなかったが,ホウキタケ科では他の種にも存在が認められた.

We examined mushrooms for inhibitors of angiotensin I-converting enzyme (ACE). We studied 23 different mushrooms that had shown ACE inhibition by preliminary screening. Of these mushrooms, marked ACE inhibitory activity was observed in <I>Rhodophyllus clypeatus, Lyophyllum semitale</I>, and <I>Pholiota adiposa.</I> Dialysis experiments showed that in mushrooms such as <I>R. clypeatus</I> and <I>L. decastes</I>, the main ACE inhibitor was a low molecular mass molecule. Conversely, the active substances in <I>P. lubrica</I> and <I>Grifola frondosa</I> were high molecular mass molecules, and ACE inhibitors from others species were assumed to contain both elements. It was demonstrated that mushrooms had specific characteristics with respect to their ACE inhibitory substances, and that these are reflected by their taxonomic affiliation at the level of family.<BR>We then isolated ACE inhibitor from <I>R. clypeatus.</I> After extraction with 70% ethanol, we obtained crystals that showed ACE inhibition and identified them to be Nicotianamine. Of the 22 other mushrooms assayed for NA content, only <I>Ramaria botrytis</I> was found to produce this metabolite.
