We studied the detection of endotoxin (ET) and peptidoglycan (PGL), as well as bacterial growth in platelet concentrates (PC) after experimental inoculation of PC bags with bacteria. ET could be detected after inoculation of 9.6×102 CFU of E. coli when the concentration increased to more than 102 CFU/ml. PGL was detected 21h after the inoculation of 4.1×10 CFU of S. aureus, and also when S. aureus concentration increased to 105CFU/ml. ET and PGL were not detected in any PC bags supplied by the Japanese Red Cross Blood Center (ET:n=161, PGL:n=210), so no side effects accompanying PC transfusion were recognized. Both ET and PGL could be measured rapidly, and detected much earlier than by the inspection of appearance. These variables may represent an index of bacterial contamination. In addition, they may be useful in analyzing the cause of side effects following the infusion of PC.
