The high sensitive, simple and new method for the determination of uric acid in serum by high-performance liquid chromatography (HPLC) is conceived and described its studies and evaluations.(1) The new method is that the pretreatment is carried out by mixing 100μl of the sample serum with 100μl of methanol and 100μl of acetonitrile containing 7-(2-hydroxyethyl)-theophylline of the internal standard which is newly selected, further the mixing solution with sodium acetate buffer (10mM, pH 4.0)/acetonitrile=9/1 is used for the mobile phase solvent. (2) Within-run reproducibility (CV=2.34〜2.99% at X^^-=4.05〜18.9mg/dl, n=10), day to day reproducibility (CV=2.48% at X^^-=9.35mg/dl) and recoveries (100.3〜108.2%) are very good. (3) Cross reactivities of 21 different species of serum coexistent substances are not recognized. (4) Correlation between HPLC method and phosphotungstate method determinations is good (n=100,r=0.998,y=1.01 x-0.149,X^^-=7.63,Y^^-=7.53). Consequently, we conclude that this method can be detected in specific and highly sensitive for the determination of uric acid in serum, utilized as a reference method for studying the determination of uric acid for drug interferences and specificities.
