In the present study, the heterogeneity of Thy-1^+ dendritic epidermal cell (Thy-1^+ DEC) was investigated by using the clones of Thy-1^+ DEC lines and original Thy-1^+ DEC. By using the antibody to deplete Ia+cell from the Thy-1^+ DEC enriched fraction, asia1oGM1-double negative cell (Thy-1^+ DEC) lines were established. The results of FACS-sorting of original Thy-1^+ DEC rich fraction showed that both asialoGM1+Thy-1^+ DEC and asialoGM1-Thy-1^+ DEC were present in this fraction. Subsequent cloning experiments revealed that the both populations derived from the double negative cell line in the Thy-1^+ DEC. By using cloned lines with the asia1oGM1+Thy-1^+ DEC from C3H mice, the heterogeneity of Thy-1^+ DEC was made clear. Especially two clones (YETY245 D3,YETY245 F45B12) of NLDC145 positive, Langer hans cell specific surface maker, were also CD3 and asialoGM1 positive. These clones had the accessory function, which implies that the clones are comparable with syngeneic or allogenic Langerhans cells. Thus the results obtained here suggest that Thy-1^+ DEC has heterogeneity, which including not only T cell lineage but also dendritic lineage.
