Staphylokinase expresses plasminogen activator activity by forming a complex with plasminogen. A cross-linked staphylokinase/plasminogen complex was produced with disuccinimidyl suberate, and its enzymatic characteristics were compared with those of a streptokinase/plasminogen complex. The staphylokinase/plasminogen and streptokinase/plasminogen complex showed a band with a molecular weight of 110kDa and 140kDa by SDS-PAGE, respectively. Both complexes exhibited plasminogen activator activity in a time- and concentration-dependent manner. FCB-2 enhanced the enzymatic activity of kinetic analysis about 5-fold for the staphylokinase/plasminogen complex, but only 1.7-fold for the streptokinase/plasminogen complex. These findings indicate that the staphylokinase/plasminogen complex binds to fibrin, and that this complex expresses plasminogen activator activity efficiently.
