Intranuclear structure represented by colloid iron stain. Comparative study with colloid iron, Ag-NORs and Unna-Pappenheim stains.

Abstract

Details of colloid iron staining of cytological specimens from ascitis or pleural effusions of 40 patients and touch smears obtained from 55 patients with malignant epithelial neoplasia were examined. In addition to the acid mucopolysaccharides, small round structures positive for the stain were found in each nucleus. Larger structures were predominant in the nuclei of malignant epithelial neoplasia cells. The shape, number and size of these structures were closely similar to those demonstrated by Ag-NORs dots as well as by Unna-Pappenheim stain. However, the stainability of these structures for both colloid iron and Ag-NORs dots was resistant to ribonuclease, but that for Unna-Pappenheim was not. The present observation suggests that the colloid iron dots demonstrate the nucleolar organizing regions in nuclei in the same way as the Ag-NORs stain. The usefulness of the colloid iron stain for revealing the details of malignant cells might be emphasized because the specimen stained with colloid iron provides not only evidence of acid mucin but the features of nucleolar organizing regions.
