遺伝子制御による異種移植の研究  [in Japanese] Organ Xenotransplantation with Gene Enginerring  [in Japanese]

Abstract

異種臓器移植は脳死ドナー不足を解決する有力な手段であるが, 移植後早期に生ずる超急性拒絶反応が最大の問題である. 我々は, in vitro で補体制御因子遺伝子導入の効果を検討するとともに, アデノウイルス法を用いて in vivo での遺伝子導入の可能性を検討した. その結果, 補体制御因子遺伝子導入により異種免疫反応は効果的に抑制されること, アデノウイルス法での遺伝子導入は特に異種移植ドナーとして最適と考えられているブタ肝臓で有効であることが明らかとなった. 以上より, 遺伝子治療の技術のひとつであるアデノウイルス法は遺伝子制御による異種移植に有効であることが示唆された.

Organ xenotransplantation is an attractive method to compensate for the cadaveric donor shortage. However, hyperacute rejection is a great barrier to successful organ xenotransplantation. In this study, the authors examined the effect of in vitro gene engineering with regulator of complement activation (RCA) molecules such as DAF and HRF20, and the possibility of applying in vivo gene engineering to the xenogeneic liver and kidney using an adenoviral vector. It was concluded that complement activation is inhibited by the gene engineering with RCA molecules against the xenogeneic cells, and that an adenoviral vector is a useful method for in vivo gene engineering to the xenogenic organs.
