Background and Aims : The present scanning electron microscope study showed the transformation of muscle capillary from the continuous capillary lacking fenestrae to the fenestrated capillary after the bilateral hindlimb casting.<BR>Methods : Five adult Wistar strain rats were killed after the bilateral hindlimb casting for two weeks. Capillary endothelial cells in the tibialis anterior muscle of hindlimb casting rats were studied by scanning electron microscopy, thin-section electron microscopy, and light microscopy.<BR>Results : Muscle fiber atrophy and enlargement of the endomysium were conspicuous in the casting rats. Muscle capillaries resided in the enlarged connective tissues. In the scanning electron microscopy, the diameter of caveolae in the casting rats was larger than those seen in the control rats, and the number of caveolae in the casting rats was greater than those seen in the control rats. Fenestrae were found only in the capillary endothelial cells of casting rats and calibrated at 76.6±28.7nm in diameter. In thin-section electron microscopy, fenestrae appeared as small gaps in the attenuated portions of the endothelial cells.<BR>Conclusions : Fenestrated capillaries in the tibialis anterior muscle appear in corresponded to the increased permeability in the endothelial cells of the casting rats.
