Abstract

The effect of thyrotropin releasing hormone (TRH) on bronchoconstriction in guinea pigs induced by leukotriene D_4 inhalation was investigated. Mean pulmonary resistance (R_L) and dynamic complaiance (C_<dyn>) were measured using a bodyplethysmograph for small animals and calculated by a microcomputor. 5 minutes following the leukotriene D_4 inhalation, C_<dyn> and R_L of the control group were -70.7±11.3% and +86.9±26.3%, those of the TRH(1mg/kg) pretreated group -60.2±11.1% and +78.8±27.8% and those of the TRH(2mg/kg) pretreated group -51.0±14.5%(p<0.05) and +81.6±27.7% respectively. However, administration of TRH(2mg/kg) 6 minutes after leukotriene D_4 inhalation exerted no influence on the effect of leukotriene D_4 on C_<dyn> and R_L. From these results, it is concluded that the sensitivity of the peripheral airway response to leukotriene D_4 was decreased by the pretreatment with TRH.
