Na-Cu-cholorophyllin (Cu-C) has been reported by Sindo et al. to be a strong complement inhibitor. Nishioka et al. have found that the reactivity of C'3b was blocked with Cu-C. Effect of Cu-C on the homograft reaction that is considered to belong to the delayed type reaction was studied. 1)Skin grafts of CFW mice transplanted in the homologous C3H/He mice survived statistically longer than controls when the recipients, C3H/He mice, were treated with intraperitoneal injections of Cu-C, 1.0mg/10gm/day, for 8 days. The 2nd-set phenomenon in this system was completely inhibited with local injections of Cu-C, 0.25mg/10gm/day, for 7 days. 2)MH134 tumor cells native in C3H/He strain inoculated intraperitoneally in the homolous C57B1 mice progressed markedly when the recipients were injected every day with Cu.C, 1.0mg/10gm/day, although the hosts resisted for a while approximate 7 days after the tumor inoculation. Thus the homograft reaction against ascites tumor was so markedly inhibited with Cu-C that the recipients died from the progressed tumor within 13 days. On the contrary, homograft reaction to MH134 tumor was accelerated when the recipients, C57B1 mice, were treated with less than approximate 0.1mg/10gm/day of Cu-C. From these results it was suggested that homograft reaction is inhibited with Cu-C markedly when target graft cells sensitized with antibody contact with concentrated Cu-C. The observations that Cu-C of the ranges of dosis employed sohwed rather stimulated antibody production to both sheep erythrocytes and tumor homograft in mice without severe toxic effects suggest that Cu-C dose not effect in the inhibition of antibody production but blocks complement reaction that causes destruction of the sensitized graft cells.
