Pre-harvest sprouting of cereal grain causes economic losses especially in cultivars. Abscisic acid (ABA) plays a crucial role in the maintenance of seed dormancy and in the prevention of pre-harvest sprouting in cereals. In this study, we performed molecular and chemical analyses to elucidate the mechanisms of germination during seed development of barley (Hordeum vurgare L.). A key regulated step in ABA biosynthesis in plants is catalyzed by 9-cis-epoxycarotenoid dioxygenase (NCED), which cleaves 9-cis-xanthophylls to xanthoxin, a precursor of ABA. Recently cytochrome P450 CYP707A gene encoding ABA 8'-hydroxylase, a key enzyme in ABA catabolism, has been isolated from Arabidopsis. Here we show the cloning of cDNAs encoding NCED and CYP707A homologs from barley, and the analysis of these expression patterns during seed development. ABA content was also studied in immature grains throughout the seed development period.
