The cell-free extract of Bifidobacterium bifidum grown on glucose, lactose, or galactose as a carbon source showed relatively high pyrophosphorylase activities towatds uridine diphosphate glucose (UDP-glucose) and uridine diphosphate galactose(UDP-galactose) and a slight activity towards uridine diphosphate xylose(UDP-xylose). No activity was found towards other sugar nucleotides.Various strains of lactic acid bacteria and actinomycetes showed little or no activities of UDP-glucose and UDP-galactose pyrophosphorylase.The activities of galactokinase, galactose-1-phosphate uridyltransferase and UDP-galactose 4-epimerasem, which are involved in the Leloir pathway, and of related nucleotide kinases, were also investigated. Activities of Leloir pathway enzymes were found in not only galactose- and lactose-grown cells but also glucose-grown cells of B. bifidum.The enzymes were separated by column chromatography on DAEA-cellulose.The activity of UCP-galactose pyrophosphorylase was one-third that of UDP-glucose pyrophosphorylase even in the DEAE-cellulose fraction free of UDP-galactose 4-epimerase and galactose 1-phosphate uridyltransferse. The existence of an additional pathway of galactose metabolism was also discussed.
