抄録

The author has developed the HPLC measurement of 6-aminonicotinamide, which is the most powerful antagonist of niacin. The analytical method for the measurement of 6-aminonicotinamide used a 7-ODS-L column (250 mm × 4.6 mm I. D. , particle size 7 μm) eluted with 10 mM potassium dihydrogenphosphate-acetonitrile (96 : 4, v/v, pH was adjusted to 3.0 by addition of concentrated phosphoric acid) at a flow-rate of 1.0 ml/min. The UV detector was set at 266 nm. The column temperature was maintained at 25℃. Under the conditions, 6-aminonicotinamide was eluted at about 4 min and the simultaneous measurements of nicotinamide, N^<-1>-methyl-2-pyridone-5-carboxamide and N^<-1>-methyl-4-pyridone-3-carboxamide were possible. The detection limit was 4 pmol at a signal-to-noise ratio of 5 : 1. The total time required for one analysis was around 15 min. The 6-aminonicotinamide in the liver and urine of the rats fed with a diet being added 6-aminonicotinamide was measured as test food samples containing 6-aminonicotinamide.
