抄録
Cytosolic adenylate kinase (AK1; [EC 2.7.4.3]) synthesizes thiamin triphosphate (TTP) both in vitro and in vivo. AK 1 synthesizes TTP by a reaction: TDP + ADP ⇄ TTP + AMP in addition to its main reaction: 2ADP ⇄ ATP + AMP. No TTP-synthesizing enzymes other than AK 1 were detected in the skeletal muscle. TDP is assumed to bind to the less-specific nucleotide binding site (ATP binding site) of AK 1 as a substrate, while ADP binds to the highly-specific site (AMP binding site), which results in formation of TTP by phosphate transfer from ADP.
