Quaternary alkaloids form an ion-pair with the perchlorate anion, which is quantitatively extractable from aqueous soltions to 1, 2-dichloroethane under acidic conditions. Many tertiary alkaloids are also extractable, though the extractability is not always high. The crude alkaloidal fraction obtained by ion-pair extraction is separated by means of ion-pair HPLC, in which perchlorate is again utilized as an ion-pair reagent. The ion-pair HPLC system containing perchlorate gives excellent separability that is comparable to conventional ion-pair systems containing alkylsulfonate or dodecyl sulfate; furtheremore, the acquisition of alkaloids is easy under the solvent system: From the fraction obtained by preparative HPLC, the organic solvent is evaporated off, and from the residual aqueous phase containing sodium perchlorate alkaloids were extracted with 1, 2 dichloroethane. The evaporation of 1, 2-dichloroethane yields pure perchlorate of the alkaloid. A few applications to several medicinal plants were performed, and from the stem of Sinomenium acutum main alkaloids, sinomenine and magnoflorine, were obtained as pure perchlorates.
