抄録

A spectrophotometric method was established for the determination of protein with pyrocatechol violet-tin(IV) {PV-Sn(IV)} complex. This method can be used to determine up to about 200 μg/10 ml of human serum albumin (HSA), and is about 2 times more sensitive than the pyrogallol red-molybdenum(VI) method (PR method). Results obtained for urinary protein by the PR method agree well with this method, the regression equation being Y (this method) =0.954X (PR method) +4.318 (n =35, r =0.986). A sensitive preconcentration method for spectrophotometry of trace protein which involves the filtration of an association complex [HSA-PV-Sn(IV)] on a membrane filter followed by its redissolution is described. The binding parameters (n and K) were calculated from a dual double-reciprocal plot. The thermodynamic parameters (ΔG, ΔH and ΔS) were obtained from a van't Hoff plot and the binding between HSA and PV-Sn(IV) complex was presumed to be a hydrophobic interaction.
