抄録

Weak bases and their salts listed in Japanese Pharmacopoeia were determined by nonaqueous titration after extraction or hydrolysis. Aim of this study was to omit the troublesome pretreatments. The drugs were determined directly by potentiometric titration with perchloric acid in acetic anhydride-glacial acetic acid (10:07:3). The titration curves, the first and second derivative curves after smoothing, were recorded on an automatic titrator assisted by a microcomputer. Sodium benzoate (Ib) and caffeine (Ia) of their mixture (I) were determined by two steps of titration curves, respectively. Imipramine hydrochloride (II) and hydroxyzine pamoate (IV) were determined by the first and second steps, respectively. Diphenhydramine tannate (III) gave a clear end point. Phenacetin (V) showed a small potential jump at the end point only on the derivative curve. Standard deviation of this method was ca. 0.3% for IIV and 0.8% for V. It was estimated from the potential of glass electrode at the half equivalent point that the order of base strength was alkylamines (II, III, IV), sodium carboxylate (Ib), amine hydrochlorides (II, IIIa), diarylmethylamine (IV), purine (Ia), amide (V) and diarylamine (II).
