Various hydroperoxides, such as cumen hydroperoxide, t-butyl hydroperoxide and ethyloleate hydroperoxide, react with new color reagents containing 4-aminoantipyrine (4-AA) or 3-methyl-2-benzothiazolinone hydrazone (MBTH), N, N-dialkylaniline derivatives and iron(II) ion, in acidic medium to yield quinoneimine type chromogens {λmax: (530601)nm}. From the results obtained by various combinations of reagents, the combinations of N-ethyl-N-(β-hydroxyethyl)-3-methylaniline and 4-AA (Reagent A) or MBTH (Reagent B) were recommended. The reagents and the assay procedure are as follows: Reagent A (or B) was prepared by dissolving 8.4 m mol of N-ethyl-N-(β-hydroxyethyl)-3-methylaniline, 1.5 mmol of 4-AA (or MBTH) and 50 mg of ferrous ammonium sulfate in 11 of 0.05 M acetate buffer (pH 4.0). A sample containing (40100)nmol of hydroperoxide was mixed with 4 ml of 2-propanol and centrifuged. One ml of reagent A was then added to 3 ml of the supernatant solution, and the mixture was allowed to stand for 60 min at 37°C. The developed colored solution was measured spectrophotometrically. When reagent B was used, 1-propanol was used instead of 2-propanol.
