Immunohistochemical Study of Thyroid Transcription Factor-1 Expression in Lung Cancers.

Abstract

Objective: Recent studies have indicated that the expression of thyroid transcription factor-1 (TTF -1) may be a usefulmarker for lung cancer. In this study we investigated the expression of TTF-1 in lung cancers of various histologicaltypes, and also evaluated the utility of TTF-1 in discriminating between pulmonary and extrapulmonary adenocarcinomas.<BR>Methods: We immunostained 140 carcinomas of pulmonary origin (60 adenocarcinomas (Ads), 30 small cell carcinomas (SCLCs), 40 squamous cell carcinomas (SCCs), 10 large cell carcinomas (LCs)), and 120 adenocarcinomas of extrapulmonaryorigin, using a monoclonal anti-TTF-1 antibody and Catalyzed Signal Amplification (CSA) system on paraffinsections.<BR>Results: In the normal lung, TTF-1 expression was restricted to the nuclei of type II alveolar epithelial cells, Clara cellsand some basal cells of the bronchioles. In lung cancers, TTF-1 expression was detected in 50 cases (83%) of 60 Ads, 20cases (67%) of 30 SCLCs, 3 cases (8%) of 40 SCCs, and 2 cases (20%) of 10 LCs. The frequency of TTF-1 expressionin pulmonary adenocarcinomas showed a positive correlation with degrees of histological differentiation. TTF-1 expressionwas found in 28 cases of 30 thyroid cancers, but otherwise in only one case of 90 extrapulmonary adenocarcinomas.<BR>Conclusion: The results suggest that TTF-1 can be a very useful marker discriminating adenocarcinomas of pulmonaryorigin from those of other sites, excluding the thyroid.
