Aims: We have previously shown that rat olfactory receptor neurons (ORNs) in the olfactory epithelium (OB) at postnatal day-one express messenger RNA of brain-derived neurotrophic factor (BDNFmRNA). The present study was conducted to examine distributional patterns of BDNFmRNA in ORNs which are present the posterior nasal cavity (PNC) at 6-week-old. Methods: The nasal cavities of seven rats were fixed with Zamboni's fixative by perfusion and 10 mm-thick frontal sections by a cryostat were prepared. The identification of the OE in the PNC was performad by immunostaining of olfactory marker protein (OMP) that labels mature ORNs. The presence of BDNFmRNA was detected by in situ hybridization histochemistry in which digoxigenin (Dig)-labeled cRNA probe and anti-Dig antibody were used. Results and Discussion: Most of the epithelium in the PNC was occupied by the OF that included OMP-immunoreactive ORNs. However, the intensity of Dig-immunoreactivity that accounts for the presence of BDNFmRNA was not homogeneous throughout the OE. For example, the OB covering the nasal turbinates tended to show strong immunoreactivity, whereas the OE covering the nasal septum showed weak immunoreactivity. These results suggest that the quantity of BDNFmRNA in ORNs are regionally different in the PNC 6-week-old rats.
