Abstract

エビ類の鮮度判定に有用な、オルニチン量を簡便・迅速に計測するバイエンザイムリアクター型センサシステムの開発を行った。本センサシステムは、オルニチンカルバミルトランスフェラーゼ(OCT)とピルビン酸オキシダーゼ(POPG)をガラスビーズに固定化して酸素電極と組合せて構成した。これにカルバミルリン酸とピルビン酸を共存させた緩衝液を通液し、オルニチンをOCTでシトルリンとリン酸イオンに分解し、このリン酸イオンを用いたPOPGの酵素反応による酸素減少量からオルニチン量を計測する。

Bienzyme reactors system was developed for the determination of ornithine. The system was prepared from the immobilized enzyme reactors (ornithine carbamyl transferase : OCT and pyruvate oxidase : POPG) and an oxygen electrode. Tris-HCl buffer solution, containing carbamyl phosphate and pyruvic acid, was continuously transferred into the system at 35°C. Phosphate ion was formed enzymatically (OCT) by transformation of ornithine in the presence of carbamyl phosphate. Pyruvate oxidase is activated by the existence of phosphate. Therefore, ornithine is determined as oxygen consumption in enzyme reaction.
