High grade lecithin (egg) was prepared by counter alternative current chromatography (CACC) with different partition between two immiscible liquids for chromatographic purification of various substances. The most distinctive feature of the CACC is a system sending alternately upper and lower phase as mobile phase to the column. Consequently, without a long size column, separation is proceeded by moving the solutes back and forthe within the column assembly. The commercial reagent grade lecithin developed more than nine spots on a silica gel thin layer chromatography (TLC) plate and was identified to be a mixture containing ca. 21% of lecithin. The effect of the numbers for switchover of mobile phase and the amount of sample on the purification of lecithin were investigated. The phase system used for the purification is composed of n-heptan-n-buthanol-chloroform-methanol-60% acetic acid (3 : 2 : 2 : 3 : 5, v/v). The preparation of high grade lecithin forming stable liposome was obtained by two or four times switchovers of the mobile phase. The present method is applicable to the preparative purification of natural products, particularly polar compounds, by using the system with liquid two phases highly efficiently.
