Few studies have dealt with in vitro culture of mangrove plants. For the establishment of in vitro culture, it is necessary to investigate the characteristics of nutrient absorption in detail. In this study, in vitro nutrient absorption in S. caseolaris and A. corniculatum was investigated. S. caseolaris grew well during the first three weeks after in vitro planting, but there after, slow growth, browning of some roots and low pH of the culture media were observed. It was found that the nutrients absorbed consisted mostly of potassium and nitrogen, mainly in the ammonium form. The absorption of nitrate was half of that of ammonium, resulting in a low pH. An identical absorption of ammonium and nitrate was observed for A. corniculatum, and the pH of the media did not decrease appreciably. When S. caseolaris and A. corniculatum were cultured on media containing nitrate in the nitrogen form alone, some chlorosis of the leaves was observed, due to nitrogen deficiency. Lack of ability to utilize nitrate was considered. Although it is well known that many plants prefer nitrate to ammonium as forms of nitrogen, S. caseolaris and A. corniculatum preferred ammonium to nitrate, and these species displayed nutrient absorption characteristics different from those of many plants, presumably because they grew under reduced soil conditions with a large amount of ammonium as nitrogen form. Forin vitro development of mangrove plants, it is necessary to take this special characteristic into consideration.
