MDSを用いた馬主席卵胞のLHとPGF_<2α>感作試験(臨床繁殖学) Effects of LH and PGF_<2α> in Equine Dominant Follicles Observed by MDS (Theriogenology)

Microdialysisシステム(MDS)は, 異なる細胞間の内分泌学的解析を分子レベルで行う方法である.今回, 馬の排卵前卵胞液中プロジェステロン(P_4), エストラジオール17β(E_2), アンドロステンジオン(A_4)とプロスラグランジンF_<2α>(PGF_<2α>)濃度を調べた.卵巣から卵胞を取り出し, ホルモン測定のため卵胞液を採取した.卵胞の大きさを大(30mm<), 中(29〜16mm)および小(15mm>)に区分けして, 各々の卵胞液中のホルモン濃度をEIA法により測定した.卵胞の大きさの違いで, P_4とPGF_<2α>濃度に有意差はなく, (A_4)濃度は大と中の卵胞間で差異を認めた.E_2濃度は, 中卵胞より大卵胞が高値であった.卵胞膜の小片にMDS操作を行い, LHを感作した時にP_4,A_4とPGF_<2α>分泌は増加した.PGF_<2α>を感作した時, P_4とA_4分泌の増加は認められたが, E_2の変動は小さかった.今回の結果から, 排卵前卵胞はLH感作により卵胞膜での黄体化とステロイドホルモン分泌を促進した.さらに, 発情周期のホルモン動態は, 他の動物種と大きく異なることが明らかとなったが, 主席卵胞が選択される機序は十分説明できなかった.

Microdialysis System (MDS) is a novel technique used for investigation of molecule secretion between different cell populations. Local hormonal secretion at follicular wall has been still unclear. This MDS study was used to determine progesterone (P_4), androstenedione (A_4), estradiol-17β (E_2) and Prostaglandin F_<2α> (PGF_<2α>) release in mare pre-ovulatory follicles. Follicles larger than 30 mm were isolated from the ovary and follicular fluid aspirated for hormone assay. Follicular fluid collected from small, middle and large follicles were analyzed by EIA. The concentrations of P_4 and PGF_<2α> were similar among the different sizes of follicles. The release of A_4 was observed in middle and large follicles. E_2 concentration was observed in middle follicles and was higher in large follicles compared with middle follicles. Follicular wall was cut and incubated for MDS and when LH was infused, there was an increase in P4 and A_4 release. PGF_<2α> release was considerably high after LH infusion compared to the control group. Infusion of PGF_<2α> increased P_4 and A_4 release but there was no change in E_2 release. This results suggest that in pre-ovulatory follicles, LH stimulates theca interna cells and also PGF_<2α> seemed to have a mediator role to induce steroid hormone production and luteinization of follicular cells. The nature of the mechanisms involved in selection of large follicles is still a perplexing research problem in reproduction.
