Qualitative analysis of midband lipoproteins, remnant and lipoprotein (a), by agarose gel electrophoresis and differential staining of lipids

Objectives : Although midband lipoprotein, a heterogeneous fraction containing very low-density lipoprotein (VLDL)-remnant, lipoprotein(a) (Lp(a) ), or modified low-density lipoprotein (LDL), promotes atherosclerosis, it is difficult to assess by conventional electrophoresis. This study aimed to assess the differences in lipid composition of midband lipoproteins. Design and Methods : Fifty subjects with midband lipoproteins detected by polyacrylamide gel electrophoresis (PAGE) were recruited and electrophoretic patterns were examined by a combined system of agarose gel electrophoresis and enzymatic stain of cholesterol (CHOL) and triglyceride (TG). Results : Subjects were divided into either a TG-rich midband group or a TG-poor midband group by a midband-CHOL/midband-TG ratio of 0.5 in this system. Lipid characteristics of the TG-rich midband group (ratio of <0.5) were similar to those of VLDL-remnant. The TG-poor midband group (ratio of >0.5) had a higher plasma Lp (a). Conclusion : This system was useful in analysis of the qualitative abnormalities of midband lipoproteins, particularly (we found) a midband-CHOL/midband-TG ratio of 0.5 may indicate the increase of VLDL-remnant or Lp (a).
