This study examined the relationships between the presence of Actinobacillus actinomycetemcomi-tans, Porphyromonas ginivalis, Prevotella inter-media, Fusobacterium nuleatum, Treponema denti-cola and Campylobacter rectus in subgingival micro-flora by using DNA probes and subsequent assess-ment of clinical response before and after scaling and rootplaning. <BR>A total of 25 subgingival plaque samples were collected from 6 patients suffering from adult peri-odontitis. The average probing pocket depth was 6. 12 mm. Subgingival plaque samples were col-lected before and at 4 and 8 weeks after scaling and rootplaning. Following determination of clinical indices after 14 weeks, all sites were classified into two groups ( the well responding group and the non -responding group ) according to their clinical responses compared to the baseline. In the well responding group, there were significant gains in the clinical attachment level (CAL) values during the entire experimental period. On the other hand, there were no significant changes in CAL values in the non-responding group during the experimental period. In addition, there was only a significant difference ( p=0.02) in the positive ratios of T denticola at week 8 between the two groups when all detectable bacterial species from all sampling periods were compared. These results suggested that the presence or absence of T. denticola among subgingival microflora at 8 weeks may predict a clinical response at 14 weeks. T denticola may therefore be a useful diagnostic marker for future clinical response after treatment, when low (103 cells<6×104) cell levels, or more, were consid-ered “positive sites”.
