コシアブラ種子の胚発育に及ぼす湿潤低温処理時期と期間およびGA_3処理時期の影響(繁殖・育種)  [in Japanese] Effects of Cold Staratification Procedure and GA_3 Treatment on Embryo Development in Acanthopanax sciadophylloides(Propagation & Transplant Production)  [in Japanese]

Abstract

コシアブラにおける種子繁殖法の確立のための発芽促進を目的として,成熟果実中から種子を採種し,GA_3処理(200mg・L^<-1>)と層積法を用いた20℃湿潤処理および5℃の湿潤低温処理を行った.パラフィン切片法による種子の組織学的観察により胚の生育ステージを調査した結果,採種直後の種子の内部は,ほとんどが胚乳で占められており,球状胚から心臓形胚までの未熟胚であった.胚の生育に対するGA_3前処理および20℃湿潤処理の効果を検討したところ,GA_3前処理+20℃4か月処理で胚の生育が促進された.20℃湿潤処理後,GA_3後処理および湿潤低温処理を行ったところ,GA_3前処理+20℃3か月処理した種子で湿潤低温処理により胚の生育が促進され,種子は発芽がみられたが,20℃4か月処理した種子では,腐敗が多く観察された.以上の結果より,GA_3前処理は20℃湿潤処理による胚の生育を促進すること,また,その後の湿潤低温処理により胚の生育はさらに進むことが明らかとなった.また,早期に実生を得るための処理としては,GA_3前後処理+20℃湿潤3か月処理+湿潤低温4か月処理が効果的であると考えられた.

To establish of seed propagation methods in Acanthopanax sciadophylloides, GA_3 application (200mg・L^<-1>), warm (20℃) stratification and cold (5℃) stratification procedures were conducted. Seeds in ripening fruits had immature embryos showing globular or heart shapes. GA_3 pretreatment accelerated embryo development among plants receiving four-months warm stratification treatment. The effect of cold stratification treatment on embryo development was observed in seeds with GA_3+three-months warm stratification treatment. After four months of warm stratification treatment, however, many seeds decayed during cold stratification. These findings indicated that GA_3 treatment prior to warm stratification treatment and following cold stratification treatment promoted embryo development. The most effective method for seed ripening was GA_3 pre- and post-treatment plus three months of warm stratification plus four months of cold stratification treatment.
