The spirochetes are composed of three main structures : the outer envelope, protoplasmic cylinder and axial fibrils (flagella). They are typical prokaryotes and are classified as bacteria since no nuclear membrane are present and muramic acid or peptidoglycan were present in the cell wall or cell wall-membrane complex of these organisms. Outer envelope polysaccharide fractions extracted by ammonium hydroxide (NH_4OH-PS) were obtained from various representative members of genera of the family Spirochaetaceae. The sugar composition of NH_4OH-PS preparations was complex and many kinds of sugar such as rhamnose, fucose, ribose, xylose, mannose, galactose and glucose were detected in all spirochetes. Of particular interest was the presence of 4-O-methylmannose as a unique constituent of polysaccharide in members of the geuns Leptospira. This sugar was not detected in the polysaccharide of Spirochaeta, Borrelia and Treponema. The chemical compositions of cell wall fractions were also examined. 4-O-Methylmannose was detected in the cell wall polysaccharides of the genus Leptospira but not in the cell walls prepared from the other genus spirochetes. The diaminopimelic acid present in cell wall peptidoglycans of the genus Leptospira was meso-diaminopimelic acid (A_2pm). In contrast to the Leptospira, the peptidoglycans of Spirochaeta, Borrelia and Treponema contained ornithine (Orn) but not A_2pm. Since 4-O-methylmannose and A_2pm were found in the cell wall fractions of genus Leptospira but not in Spirochaeta, Borrelia or Treponema, it was suggested that the chemical composition of the cell wall becomes an important criterion of chemotaxonomy of Spirochaetales.
