Intracellular calcium elevation (calcium wave) is initiated by various stimulations. Moreover, when single cell is stimulated, intracellular calcium wave propagates to the neighboring cells. These factors indicate that endothelial cells passes signal between cells through intercellular calcium wave propagation. Recent studies of calcium wave propagation had shown that one of calcium wave propagation pathway is ATP production from stimulated cells. On the other hand, there is still little knowledge on intracellular signal transduction when intercellular calcium wave happens. We investigated the location of Protein kinase C alpha (PKCalpha) which is one of a calcium sensing protein using fusion protein PKCalpha-AcGFP in intercellular calcium wave initiated by point mechanical stimulation. As the result, ATP stimulation changed PKCalpha intracellular location from cytoplasm to plasma membrane. Moreover point mechanical stimulation initiated intercellular calcium wave propagation and changed PKCalpha intracellular location of stimulated cell and unstimulated cells.
