A new in vitro embryotoxicity test is demanded to examine the effects of agents on human metabolic activity. We investigated the hybrid culture of mouse ES cells and three hepatocyte lines according to a 3-D culture protocol with glass fiber. The hepatocyte lines are human and mouse floating cell lines and HepG2. No hepatocyte line was used for a control group. The effects of thalidomide, nonteratogenic in mice, were examined. Thalidomide influenced the metabolic activation of the floating hepatocyte lines as compared with the control group. The protocol will be improved in the future.
