Inhibition of P-glycoprotein by multi-walled carbon nanotubes in Caco-2 cells

CaCo-2 cells with constitutive P-glycoprotein (Pgp) expression were exposed to multiwalled carbon nanotubes (MWCNTs). Subsequent flow cytometry showed decreased efflux of intracellular Pgp substrates. RT-PCR and Western blotting analysis showed reduced Pgp mRNA and protein respectively. We conclude that MWCNTs could decrease Pgp expression both at transcriptional and translational level, and thereby reducing Pgp-mediated drug transport.
