We have reported that OCP-gelatin (OCP/Gel) composites have a potential to enhance bone formation in various bone sites. However, certain mechanical stress suppresses the bone formation property of OCP under in vivo condition. Mechanical stimulation is considered to be one of the significant factors regulating the homeostasis, especially, skeletal structure and function. In the present study, we developed a cyclic mechanical loading device to evaluate the effect of mechanical stress on the osteoblastic differentiation of stem cells seeded on the three-dimensional scaffold. The effect of compression was analyzed using mouse mesenchymal stem cell line D1 cells by deforming the cell-seeded OCP/Gel composite. The results showed that osteoblastic differentiation of D1 cells was promoted if suitable conditions regarding mechanical stress were provided.
