抄録
Mitochondrial behaviors and functions are prone to be affected by external forces applied through cell cytoskeleton. To investigate mitochondrial responses against applied mechanical forces, we developed a laser confocal microscope with a bent optical probe that is able to measure mechanical and physiological properties of mitochondria simultaneously. The repetitive transient depolarizations of the inner mitochondrial membrane induced by proton pumping were successfully observed. And, the mitochondrial Young's modulus was also measured by indentation experiment. Simultaneously, when applying a 30 nN on the mitochondrion, it was found that the period of depolarization increases to be 1.7 times of the normal mitochondrion.
