During colonization of the gastrointestinal tract, enterobacteria construct a hierarchic regulatory network of genome expression to adapt to environmental stresses. My studies revealed novel cascade regulations by YdeO and GadE, which are transcriptional factors known to control the glutamate-dependent acid resistance system in Escherichia coli. YdeO forms the characterized two transcriptional cascades, YdeO > YiiS > CadC and YdeO > GadE for controlling two acid resistance systems, which are dependent on glutamate and lysine. Moreover, GadE induced by YdeO forms other regulatory cascades, GadE > HdeD > LrhA > FlhDC and GadE > HdeD > GadX. Taking all the results together, my thesis presents a hierarchic multi-regulatory network followed by acid resistance factors to achieve colonization in a large intestine within host animals.
