A colorimetric method was described for the quantitative determination of lactose in milk, based on the reaction of lactose with the Somogyi's 1952 alkaline copper reagent in combination with the Nelson's arsenomolybdate reagent. After deproteinization with basic zinc carbonate or sodium tungstate and alum, the copper reagent was added to the clear solution containing 0.1-0.6mg/ml of lactose. The mixture was heated at 100C for 40min and cooled. The optical density of the colored solution was measured at 500nm within 4hr after the addition of the Nelson's reagent. Recovery (lactose found/lactose expected) of lactose in milk from fortified samples averaged over 99.6%. The precision of the method used sodium tungstate and alum as the deproteinizing agent was good with a coefficient of variation of 0.59 for replicate analyses. Comparative results among the present, the Lane-Eynon and chloramine-T methods were discussed.
