The hypothalamo-neurohypophysial neurosecretory system (HNS) and the adenohypophysis were observed for histological changes following adrenalectomy and thermal treatment from a neurosecretory point of view. It was made clear that adrenalectomy was followed by a temporary phase of increased secretory activity in both supraoptic and paraventricular nuclei at the end of post-operation (p-o) day 1. Such occasional hyper-activity, however, was gradually replaced by the normal behavior in the course of p-o days 3, 5, and 8. On p-o day 1, the acidophils of the adenohypophysis increased greatly in size and number, but decreased gradually in number in the course of p-o days 3 and 5. On the contrary, the basophils and chromophobes increased greatly in number at the end of p-o day 8.
Low-thermal treatment (in which rabbits had been transferred from room temperature at 30°C to an ice-box at 5°C) augmented the production and release of neurosecretory granules in the HNS 10 and 20 minutes after treatment. Then the neurosecretory activity was replaced by the normal moderate behavior 30 minutes after treatment. In all cases subjected to this treatment, the acidophils of the adenohypophysis increased greatly in number. The same augmentation of neurosecretory activity could be seen in the HNS after high-thermal treatment(in which rabbits had been transferred from room temperature at 30°C to an incubator at 60°C). In 10 minutes after this treatment, the acidophils of the adenohypophysis increased in number. On the contrary, 20 minutes after this treatment, the basophils of the adenohypophysis increase. In animals under thermal treatment, degranulation frequently could be seen in both acidophils and basophils of the adenohypophysis.
From the results mentioned above, it was postulated that adrenalectomy and thermal treatment provoked an elevation of neurosecretory activity in the initial period of treatment and that the increase of neurosecretory substance in the adenohypophysis was followed by an increase of acidophils and stimulated a release of granules of both acidophils and basophils.
