A monoclonal antibody against the low-sulfur (LS) component of bovine hair keratin was prepared. Immunoblot analysis revealed that this antibody reacted with 60 kilodalton, 50 kilodalton and 45 kilodalton keratin polypeptides of LS component but not with 70 kilodalton polypeptide of LS component nor with high-sulfur component. By immunoperoxidase staining, this antibody exclusively reacted with hair components including outer root sheath epithelia. Cells in the cuticle, the cortex and the medulla at the deepest zone of the hair follicle were well stained with this antibody, and the reactivity with the cuticle and the cortex was diminished according to karatinization, while no positive reaction was observed in the hair matrix cells and the epidermis. This antibody, in addition, stained the outer root sheath epithelia at the deeper zone than at the zone where hair goes through the final hardening well. But, at the region near the epidermis, no positive reaction with the outer root sheath cells which were not keratinized could be detected, suggesting that these cells express a new type of keratin molecule instead of the hair-type keratin as a cytoskeleton. The antibody obtained in this study, therefore, is a useful marker for investigating keratin expression during hair development and the switching mechanism of the keratin molecule expressed in the outer root sheath epithelia.
