The restriction enzyme cleavage patterns of mitochondrial DNA of the RR mouse strain is different from those of C57 BL/6 and BALB/c. Using these inbred strains and the lines of which mitochondrial DNA was substituted for that of different types by successive backcrossing for ten generations between RR and the other two strains, the maternal inheritance of enzyme activity of the mitochondrial respiratory chain was investigated. It was confirmed that the cytochrome c oxidase (complex IV) activity of RR was lower than those of the others and some of the differences were maternally inherited according to analyses of fresh and sonicated mitochondria and submitochondrial particles. NADH-ferricyanide reductase (complex I) activity was higher in RR than in C57 BL/6 and BALB/c, but strain differences were not maternally inherited. There were no strain differences in the activities of rotenone-sensitive NADHcytochrome c reductase (complex I and III) and succinate-cytochrome c reductase (complex II and III). The activity of rotenone-sensitive NADH oxidase, which measures the entire respiratory chain from NADH to O2, was associated with cytochrome c oxidase activity and lower in RR than in the other two strains. The maternal inheritance of the activity was observed in the experiment using RR, C57 BL/6 and their backcross lines, but it was not evident in another experiment using RR, BALB/c and their backcross lines.
