The phagocytic activity and the morphological changes of the Kupffer cells have been studied on the mice which received the intraperitoneal inoculation of Mouse Hepatitis Virus-2. The phagocytic activity has examined by the carbon clearance method. After the inoculation, the phagocytic activity continued to increase for 36 hours. However, the phagocytic activity began to decrease from 48th hour. The decrease became very marked after the 60th to the 96th hour. The statistical, positive correlation was established between the phagocytic activity and the count of the Kupffer cells with phagocytized carbon particles in both the midzonal and peripheral areas of the hepatic lobules. Light-microscopically, the proliferation and contrariwise also the degeneration and necrosis of the Kupffer cells were observed before the occurrence of the hepatic parenchymal necrosis. The focal necrosis of the liver cells was revealed at the 48th hour after the inoculation. The submassive or massive necrosis followed on the focal one. The virus-like particles were electrn-microscopically found within the Kupffer cells from the 6th hour after the inoculation. Furthermore, the mortality was significantly exacerbated by the blockade of the RES preceding the inoculation of the virus in comparison with the inoculation without the blockade. Necrosis of the liver cells was also accelerated by the blockade.
